Effect of substrate elasticity on TGF-β receptor and TGF-β expression by cultured renal tubule cells. TGF-β and TGF-β receptor expression levels were determined by qPCR in primary HREC cultured on soft (0.5 kPa) and stiff (50 kPa) hydrogels. TGF-B1R1 and R2 mRNA increased with increasing substrate stiffness from 0.5 to 40 kPa (A). However, TGF-β mRNA did not vary significantly as a function of substrate elasticity (B). The gene expression level was shown as the relative fold comparison with that from cells on 0.5 kPa gel, which was set as 1. *p < 0.05, **p < 0.01. The results were calculated from three independent experiments.
