Working model of the modulation of D2R signaling by NCAM. The transmembrane proteins NCAM and D2R are expressed at the cell surface of the postsynaptic terminus (1). On DA stimulation, cell surface NCAM forms a complex with D2R by interaction between intracellular domains of the two molecules (2), promotes D2R internalization (3) to endocytic compartments, and leads to the attenuation of D2R-mediated signal transduction (4).
