Arginase I and polyamines are upregulated by a peripheral, conditioning lesion. A, At various times, as indicated, after a unilateral, peripheral lesion to the sciatic nerve the DRGs were removed and subjected to Western blotting and stained for Arg I (I). The contralateral, uninjured DRGs were used as a control (C). Total protein at 50 μg were loaded per well. Blots were stripped and reprobed for actin as a loading control. B, DRGs were removed at 2 d (striped bars), 4 d (white bars), or 7 d (confetti bars) after a peripheral lesion or unlesioned (black bars) and cultured overnight before the cells were lysed and subjected to HPLC analysis for the polyamines putrescine (PUT), spermidine (SPD), and spermine (SPM).
