HAdV-5 binds to murine serum components in a FX-independent manner, but in a complement-dependent way. (A) HAdV-5 binds to immunocompetent murine serum components regardless of the presence of FX. Alexa Fluor 532-labelled HAdV-5 was incubated at room temperature for 90 min in murine sera -/+ X-bp. Samples were diluted in PBS and injected into the Nanosight LM14. Data from a minimum of three separate experiments with ~700 completed tracks are shown as the mean ± SEM. One-way ANOVA and Tukey’s post hoc test applied. * p < 0.05 vs. matched control. (B) Significant particle size drop was observed incubating HAdV-5 in heat-inactivated immunocompetent murine serum compared to active serum. Alexa Fluor 532-labelled HAdV-5 was incubated at room temperature for 90 min in murine sera. Serum inactivation was carried out by a 30 min 56 °C incubation.
