VSV binding and entry is more efficient in rU87 glioma cells. Quantification of VSV by RT-PCR at different intervals in distinct cellular compartments indicated that VSVrp30a binds and enters the rU87 human glioma cell line more efficiently than its normal cellular counterpart. Cell-surface-bound VSV after 20 min of incubation with the virus was significantly greater in rU87 cells compared with normal human astrocytes (A). When surface-bound plus internalized virus was quantified at 30 min, there was a trend toward more virus in rU87 but the difference was not significant (B). At 90 min after infection, there was significantly more VSV in the endosomal compartment in rU87 cells (C) when compared with astrocytes. Also the total amount of intracellular virus was significantly greater in rU87 cells (D). All experiments were done in triplicate using VSVrp30a at an MOI of 10.
