Figure 6. — The p53 I255del mutant exhibits reduced transcriptional activity. Constructs encoding p53 wt (TP53-wt), p53-R175H (TP53-R175H), and p53-I255del (TP53-I255del) were transiently transfected into p53-deficient Saos-2 cells. Transcriptional activity was assessed using a promoter–reporter assay. (A) The relative p53 luciferase activity was significantly reduced in cells transfected with TP53-I255del as compared to TP53-wt (P < 0.001) and was like that observed in cells transfected with TP53-R175H. (B) Corresponding p53 protein expression was determined by western blot analysis. Data represent two independent experiments, with each experiment comprising two biologic replicates.
