Effect of IFN-γ on oligodendrocytes. Mature oligodendrocyte cultures were treated for 2 d with IFN-γ (0–100 U/ml), and several cellular effects were measured. A, IFN-γ and oligodendrocyte viability. IFN-γ (100 U/ml) had no effect on oligodendrocyte viability; potentiated the toxicity of anti-fas, staurosporine, and ceramide; and protected oligodendrocytes against H2O2 and lactacystine (*p < 0.05). B, IFN-γ protection against H2O2. Maximum protection was found with 25–50 U/ml at 50 μm H2O2. C, IFN-γ induction of proteasome activity. Maximum induction was found with 25 U/ml. D, IFN-γ protection against lactacystin. Significant protection was found with all doses of IFN-γ (25–100 U/ml) against 1 μm (*p < 0.05) and only with 25 U/ml against 10 μm lactacystine (**p < 0.05).
