Reduced interaction of PSD-95 and GKAP by 2-bromopalmitate treatment. A, Immunoblotting of proteins extracted from primary hippocampal neurons treated with 2-bromopalmitate (BrPal). After mild extraction of cells with NP-40 buffer and protein (pt) fractionation, PSD-95 was primarily in the pellet. Incubation with 2-bromopalmitate increased the amount of PSD-95 in the supernatant (sp). B, Immunoprecipitation (i.p.) of PSD-95 and GKAP from neurons treated with 2-bromopalmitate. Extraction of primary neurons with buffer containing SDS (radioimmunoprecipitation assay buffer) increased the solubility of PSD-95 in control condition. After immunoprecipitation with anti-GKAP antibody, the reduced amount of PSD-95 was observed in samples treated with 2-bromopalmitate.
