Anti-MDSC activity of CA is TLR4-dependent. The cells isolated from the spleens of TLR4−/− mice bearing MC38 tumors were seeded in a 96-well microplate (1×106 cells/well). Following treatment with different concentrations (0, 1, 2 and 4 µg/ml) of CA and DMSO for 36 h, the cells were stained with fluorescent antibodies and subjected to fluorescence-activated cell sorting analysis on a flow cytometer. (A) The data were analyzed using FlowJo 7.6 (B), and a bar chart was generated with GraphPad Prism 5.0. The data are expressed as the means ± SD (n=4), and the significance was determined by one-way ANOVA. CA, cinnamaldehyde; DMSO, dimethyl sulfoxide; NS, not significant.
