Tau modulates MB:PMCA interaction in purified pig brain PMCA. Panel A: Results of titration with MB of PMCA reconstituted in PS in the absence of tau () and after incubation for 2 min at 25 °C with 0.1 nM () or 7.5 nM tau (). Panel B: Results of titration with MB of PMCA reconstituted in PC in the absence of tau () and after incubation for 2 min at 25 °C with 7.5 nM (⬤) or 100 nM tau (). Assays were performed as indicated in the Materials and Methods in the medium used for PMCA activity measurements, but in the absence of pyruvate kinase, lactate dehydrogenase, phosphoenolpyruvate, and NADH. The inner filter effect due to the MB absorbance at 290 and 340 nm has been corrected as indicated in the Materials and Methods. Data shown are means ± SE of titration data of three experiments performed with three preparations.
