Endosulfan inhibited proteasomal activity and triggered HMW ubiquitinated protein aggregation in N27 cells. (A) N27 cells were exposed to various concentrations of endosulfan ranging from 40 to 80 μM for 20 h. (B) Cells were exposed to 60 μM endosulfan for 3–15 h. Chymotrypsin-like proteasome activity was measured using the fluorogenic substrate Suc-LLVY-AMC (75 μM). Asterisks (***, p <0.001) indicate statistically significant differences compared with vehicle-treated control cells. (C) Accumulation of HMW ubiquitin-conjugated protein in N27 cells exposed to 60 μM endosulfan. Equal amounts of proteins were resolved on 10% SDS poly-acrylamide gel electrophoresis and blotted with ubiquitin antibody.
