Generation of a Phlpp1-floxed allele. A, LoxP sites were added to the intronic sequence surrounding Exon 4 of the Phlpp1 gene via a TALEN-mediated approach. Cre-mediated recombination of the floxed allele generates a premature stop codon. Sequencing confirmed the presence of both LoxP sites and that no mutations were generated within Exon 4. Genotyping primer locations are noted (FP, forward primer; RP, reverse primer). B, genomic DNA was isolated from mature (day 4) osteoclasts derived from Phlpp1^fl/+: Cre⁻, Phlpp1^fl/+: Cre⁺ and Cre⁺ mice and assessed by PCR.
