Resupply of zwitterionic phospholipids and additives improve the stability of the inverted LacY in DDM micelles. Representative unfolding curves at 45 °C for 150 min for LacY C154G/H205C purified from PE-lacking cells and solubilized in DDM. LacY was diluted to 26 μg/mL in 50 mM Tris-HCl at pH 7 (left panels) or pH 8 (right panels) and in the presence of 150 mM NaCl (no additives, top panels) or 250 mM NaCl and 10% glycerol (NaCl + Glycerol, right panels). Resupply of phospholipids was conducted prior to thermostability assessment by incubation with DDM-solubilized phospholipids.
