(A) NLRP3 confocal analysis in isolated liver macrophages with or without HIV-1BaL infection 8 hr posttreatment of 5 μM ATP. IL-1β (magenta), gp120 (red), CD68+ liver macrophages (green), NLRP3 (orange), and nucleus (blue). (B) NLRP3 (P = 0.019), intracellular caspase-1(P = 0.039) and cleaved caspase-1 proteins in the supernatant were measured after HIV-1, LPS or HIV-1 + LPS, GAPDH was taken as internal control. The results were obtained from 8 donors. (C) Liver macrophages were infected by HIV-1BaL at an MOI of 0.10 with or without 1.0 nM MCC9450. After 100 ng/ml LPS challenge, the supernatant was collected 8 hr posttreatment with 5 nM ATP and IL-1β was measured by ELISA (n = 9, duplicate, P = 0.013).
