FIG 2

Overexpression of sRNA-29 causes nitrite accumulation and a reduction in the amount of NO produced by P. denitrificans as a consequence of reducing the expression of denitrification genes. P. denitrificans plus empty pLMB509 vector (solid line) or sRNA-29 (dashed line) was grown under denitrification conditions (20 mM nitrate as electron acceptor) for 8 h, and then 10 mM taurine was added (indicated with an arrow) to induce expression of sRNA-29 and levels of growth and denitrification intermediates were measured. (A) Optical density (OD600), (B) Nitrate (expressed in millimoles). (C) Nitrite (expressed in millimoles). (D) NO (expressed in nanomoles). (E) qRT-PCR validation of changes in expression of genes involved in denitrification. Error bars represent SD of results obtained between triplicate experiments; where not visible, the error bars were smaller than the symbols.
