AZD1775 is Synergistic with Cisplatin and 5-FU. (A) KYSE150 and EC109 cells were treated with a serially diluted mixture (a fixed ratio) of AZD1775 and CDDP or 5-FU for 72 h; the cell viability was examined by MTT assay. Synergistic effect was estimated using the median-effect method of Chou and Talalay. The combination index (CI) was the ratio of the combination dose to the sum of the single-agent doses at an isoeffective level. The dashed dotted line indicates CI = 1. CI < 1 indicate synergism between the two drugs. (B) and (D) KYSE150 and EC109 cells were exposed to AZD1775 (0.25 μM) with combination of 250 μM 5-FU or 10 μM CDDP for 48 h and then examined with a hemocytometer by trypan blue exclusion assay (n = 6 per group).
