Effects of Gomisin M2 (G.M2) on mast cell degranulation. (A) Chemical structure of G.M2. (B) Mouse bone marrow derived mast cells (BMMCs), rat basophilic leukemia cells (RBL-2H3), and rat peritoneal mast cells (RPMCs) (2 × 104 cells/well in a 96-well plate) were pre-treated with or without G.M2 and then incubated with MTT. The absorbance was detected using a spectrophotometer. For mast cell degranulation, anti-DNP (50 ng/ml)-sensitized mBMMCs, RBL-2H3 (5 × 105 cells/well in 12-well plates), and RPMCs (3 × 104 cells/well in a 24-well plate) were pre-treated with or without G.M2 or Dexa for 1 h and then challenged with DNP-HSA (100 ng/ml). (C) Histamine levels were detected with a fluorescence plate reader. (D) The level of β-hexosaminidase was measured using β-hexosaminidase substrate buffer.
