miR-130a enhances the sensitivity to irradiation of rectal cancer cells. (A) Relative expression of miR-130a was compared in SNU70RR cells with miRNA-specific real time PCR. The data were normalized with the RNU6B gene as the internal control. (B) Representative images of colony formation in SNU70RR-scramble and SNU70RR-miR-130a cells after exposure to different irradiation doses. Irradiated cells were maintained for 14 days to form the colonies and stained with crystal violet. (C) The graph shows survival fractions of SNU70RR-scramble and SNU70RR-miR-130a cells were calculated from colony formation in (B). (D) Expression of γH2AX was detected in SNU70RR-miR-130a cells after irradiation treatment for 24 h with immunofluorescence analysis. The bar graph shows the average number of γH2AX positive cells per field. Scale bar: 20 μm. The data were presented with the mean ± S.D. of three independent experiments. (⁎⁎P < .01, ⁎⁎⁎P < .001).
