Figure 6

(A) Gene structures of MTDH, YAP-1 and c-MYC showing the predicted target site of miR-375. Oncogenes regulation by miR-375. (B) Luciferase report assay in PLC/PRF/5 EveR and JHH-6 EveR cells showing that c-MYC is target of miR-375. The data presented are mean ± SEM of three independent experiments. ****p < 0.0001; ***p < 0.001. (C) VitD reduces MTDH, YAP-1 and c-MYC protein expression after 6 days of treatment in PLC/PRF/5 EveR and JHH-6 EveR cells. (D) Inhibition of miR-375 induced MTDH, YAP-1 and c-MYC protein up-regulation in both EveR cells. The same proteins, instead, were down-regulated in presence of VitD and vehicles. Conversely, MTDH, YAP-1 and c-MYC proteins were up-regulated in presence of both miR-375 inhibitor and VitD. These effects are particularly visible in PLC/PRF/5 EveR cells.
