Scheme 1 — Schematic representation of MTX-conjugated polymer hybrid micelles co-loaded with miR-124 (M-PHMs/miR-124) preparation and combination therapy of M-PHMs/miR-124 in RA. (A) The M-PHMs were formed by the self-assembly of MTX-PEI-LA and mPEG-LA. miR-124 was then loaded through electrostatic interactions with the micelles. (B) M-PHMs/miR-124 targeted to the folate receptor (FR) on activated macrophages in RA joints and released MTX conjugates and miR-124 into the cytoplasm by the proton-sponge effect. The combination therapy had both anti-inflammatory effect and joint protective effect.
