A. Positions and frequencies of template-mismatched nucleotides within Pol IV and RDR2 transcripts generated using M13 template DNA. Transcripts of 15-80 nt were examined. The frequency of template-mismatched nucleotides at each position is denoted in shades of orange/red. Related information is provided in Figure S6.

B. Consensus sequences for unique Pol IV and RDR2 transcripts longer than 26 nt. The 7 nt nearest the 5’- and 3’ ends are shown. Redundant sequences were condensed into unique sequences prior to analysis. Sequence logos were generated using WebLogo 3.6. In the summary cartoon, N denotes the untemplated nucleotide ascribed to RDR2 terminal transferase activity.

C. Sequence logos for Pol IV and RDR2 strands of 23 and 24 nt DCL3-diced siRNAs. The summary cartoon proposes a model to account for 23 nt siRNAs with 3’ terminal untemplated nucleotides.
