After the PARylation reaction, SAM and biotinylated HeLa oligonucleosomes were allowed to bind to NSD2. Streptavidin magnetic beads were added into the reaction and incubated for 1 h at room temperature. B, biotinylated nucleosomes along with bound NSD2 were captured and analyzed by immunoblot against NSD2, histone H3K36me2, and total histone H4.
