Coiled-coil domain is required for EFhd2 liquid–liquid phase separation. (A) Wild type human EFhd2 (hEFhd2WT), N-terminal (hEFhd2ΔNT), and coiled-coil (hEFhd2ΔCC) truncated mutants ALEXA-labeled recombinant proteins (4 μM) were subjected to liquid–liquid phase separation in absence (–PEG) or presence (+PEG) of 10% Polyethylene glycol (PEG) as crowding agent plus or minus 10 mM CaCl2-Recombinant tau (4 μM) was subjected to the same conditions as EFhd2. (B) EFhd2 wild type (WT), N-terminal (ΔNT), and coiled-coil (ΔCC) truncation mutant proteins and Tau proteins were resolved in SDS-PAGED and visualized by Coomassie blue staining.
