EFhd2 affects tau liquid–liquid phase separation. (A) Wild type human EFhd2 (hEFhd2WT) recombinant protein (4 μM) and equimolar concentration of recombinant tau (4 μM) were subjected to liquid–liquid phase separation in absence (–PEG; not shown) or presence (+PEG) of 10% Polyethylene glycol (PEG) as crowding agent plus or minus 10 mM CaCl2- The images are representative of, at least, three independent experiments. Scale bar = 10 μm. (B) Co-incubated EFhd2 wild type protein (4 μM) and tau (4 μM) in the presence or absence of 10 mM CaCl2 was subjected to FRAP. The intensity was normalized to the first time point before photo-bleaching and shown as the mean and standard error of the mean (SEM) through time, n = 11.
