Loss of TbL11 disrupts stages of 25/28S and 5.8S rRNA processing. Following 0 to 4 days of TbL11 RNA interference induction, total RNA was extracted from cells, stained with methylene blue, and then probed for various mature rRNAs (A) or rRNA processing intermediates (C) as labeled. Log2 values from ratios of precursor to mature rRNAs on day 0 were substracted from the corresponding log2 values on day 2 postinduction (C). Panel B shows a simple schematic of rRNA processing showing the proposed sites of processing interrupted by loss of TbL11 (43). Analyses were performed on three biological replicates, with representative blots shown.
