Time course of Gln3-Myc¹³ phosphorylation in glutamine-grown wild type and sit4Δ cells. Wild type (TB123) and sit4Δ mutant (TB136-2a) cells were cultured in YNB-glutamine medium to A_600nm =∼0.5. At that time the cells were gently collected on a Millipore filter, washed twice, and transferred to nitrogen-free YNB medium as described in Materials and Methods. Samples were then collected at the times indicated, and processed for western blot analysis.
