(A) Western blotting showing the expression of receptors in a time-dependent manner following treatment with His-MDA-7/IL-24 protein. DU-145 cells were treated with His-tagged MDA-7/IL-24 protein for the times indicated; biotinylated membrane fractions were isolated and probed with the indicated antibodies. Na-K-ATPase and EGFR expressions were used as control. Quantification of the Western blots with respect to Na-K-ATPase are as shown for IL-20R1 (B), IL-22R1 (C), and IL-20R2 (D). DU-145 cells were treated with His-tagged MDA-7/IL-24 protein for the times indicated. The cells were stained with primary antibodies followed by Alexa fluor-488 tagged secondary antibodies and flow cytometry was done to study IL-20R1 (E), IL-22R1 (F), and IL-20R2 (G) expression.
