Schematic representation of peptide-based mechanism of SMR inhibition. PAsmr homolog EmrE is depicted in a top view in its functional, dimeric form (antiparallel monomers rendered in blue and black) (PDB code 3B5D [14]). Upon the addition of PAsmrTM4 peptide (red), peptide binds antiparallel to the SMR monomer (blue), leading to disruption of the six-helix substrate transport pathway and concomitant SMR inactivation.
