GFP tagged HHD hSPCA1a variants localizes correctly to the secretory pathway in wt yeast strain PAP1500. Live cell bioimaging of yeast strain PAP1500 expressing C-terminally tagged hSPCA1a variants, the endogenous Pmr1 or Pmc1 from the replicative vector pPAP4997 at 15 °C, 20 °C, 25 °C, 30 °C or 35 °C. Yeast cultures were grown as described in Fig. 9. All images were taken at 1000x magnification. Left part of the figure shows GFP fluorescence while the right side shows the same cells visualized with differential interference contrast (DIC). The single images shown represent one out of at least five.
