Inflammatory factors inhibit ZIKV infection in a dose-dependent manner. (A to D) SNB-19 cells were inoculated with ZIKV (MOI = 0.1) with different concentrations of IL-6, IL-8, TNF-α, or MCP-1 for 48 h. Total cellular RNA was isolated, and ZIKV RNA copies were measured by qRT-PCR. (E) Antibody neutralization assay. SNB-19 cells were inoculated with ZIKV (MOI = 0.1) for 6 h, and the cells were then washed and treated with various concentrations of the antibody cocktail for another 48 h: +, 0.04 mg/liter; ++, 0.2 mg/liter; +++, 1 mg/liter. Intracellular viral RNA was measured by qRT-PCR. Isotype mouse IgG was used as a negative control. (F to I) SNB-19 cell viability was determined by MTT assay after 48 h of incubation with different concentrations of IL-6 (F), IL-8 (G), TNF-α (H), and MCP-1 (I). Values represent the mean ± SEM from independent experiments. *, P < 0.05; **, P < 0.01; ***, P < 0.001.
