ORF42 affects total protein synthesis. Protein synthesis rates were measured using AHA-based metabolic labeling. The lytic cycle was induced in the indicated KSHV-infected iSLK.RTA cells by addition of doxycycline (1 μg/mL) to the media. Five days after lytic cycle induction, nascent proteins were labeled using AHA coupled to a fluorescent dye. The fluorescence intensity of AHA-treated cells was measured by flow cytometry. The distribution of fluorescence in the cells is plotted in (a) for a representative replicate.
