Dose- and time-dependent studies showing down-regulation of IL-6 and IL-12p40 by encapsulated and naked IL-10 in mouse J774 macrophages. Macrophages were stimulated with rMOMP and incubated for over a 72 h period with different concentrations of PLA-PEG-IL-10 (1 to 100 ng/mL) and PLA-PEG-PBS (an equivalent nanoparticle weight). At each time-point, namely 24, 48, and 72 h, cytokines were quantified in cell-free supernatants by specific cytokine ELISAs for IL-6 (A–C) and IL-12 p40 (D–F), respectively. p values were calculated by the use of one-way ANOVA, followed by Turkey’s Post-test using GraphPad Prism 6 Software.
