Tissue-specific expression of TTK in 18-d-old transgenic brain tissue. In situ hybridization with TTK-Dig-labeled probes were used to detect TTK mRNA-positive cells in corpus callosum (A) and cerebral cortex (B). Immunohistofluorescence with a rabbit polyclonal antibody to HSV1-TK was performed to localize TTK-positive cells. Numerous TTK-positive cells were detected in corpus callosum (C, D) in transgenic mouse. No TTK immunoreaction was detected in control nontransgenic mouse (E). Double-labeling immunohistofluorescence shows staining of a transgenic brain section at P7 obtained with anti-HSV1-TK antibody detected by Alexa 546 (red)-tagged secondary antibody (F) and a mouse monoclonal antibody to MBP revealed by Alexa 488 (green)-labeled secondary antibody (G). Arrows show that TTK+ cells (F) are MBP+ (G) as confirmed in H. F and G are merged in H. Scale bar, 40 μm.
