Single stranded DNA examined for nick formation in neurons treated by nerve growth factors and NAP. Alkaline gel electrophoresis of DNA was used to examine nicks in single-stranded DNA. DNA was extracted from nuclei of rat brain cortical neurons, untreated (lane 1), or incubated (5 min, 25°C) with 100 ng/ml NGF, BDNF, or NT-3 and with 10-12 m NAP, as indicated. As a positive control for nicked DNA, DNA was treated with DNase I. Isolated nuclei of cortical neurons were incubated for 5 min at 37°C with DNase I (10 U; RNase free; D7291; Sigma). Single-stranded DNA in the samples was stained with ethidium bromide (1 μg/ml) and photographed under UV illumination. MW, Molecular weight.
