iNKT cells alter LDC surface molecule expression after infection. Both KO as well as WT mice (3 in each group) were sacrificed at day 3 after intranasal infection (3 × 106 IFUs/mouse). Lungs were harvested and processed into single-cell suspensions. CD11c+ lung cells were isolated by using CD11c-magnetic microbead selection. CD11chi non-autofluorescent lung cells were gated as LDCs as shown in figure 1a. The LDCs were analyzed for the expression of MHC-II and costimulatory molecules by flow cytometry. Flow cytometric dotplots are shown. Expressions of CD80, CD86, CD40 and MHC-II on LDCs (shaded histogram) and isotype control (line) were shown. The mean fluorescence intensity (left) and percentages of positive cells (right) were indicated. One of the 2 independent experiments with similar results is shown.
