Graph showing how fluorescent intensity (measured as mean pixel intensity along a line through the image) of dot blots of extracts from cerebellar tissue labeled for GLAST varies with relative GLAST concentration, expressed as a percentage of the stock solution. Mean values were obtained by combining data from extracts from four animals. For each of the four sample series, the confocal settings were adjusted to give a mean pixel intensity of ∼70 for the 100% dot, and then all subsequent dots of the series were imaged under the same conditions. With the exception of the 10, 20, and 100% dots, the values vary linearly with relative solution strength. (Filled squares represent those values that lie in the linear range; filled diamonds represent those that lie outside it.)
