C6 cells were preincubated with BIS II (10 μm) or Gö6976 (10 μm) for 10 min before the addition of PMA (100 nm) for 30 min. EAAC1 immunoprecipitates (IP) were analyzed for PKCα immunoreactivity (WB: PKCα). B, EAAC1–PKCα association is not induced by PDGF. C6 cells were incubated for 30 min with PDGF (20 ng/ml) or PMA (100 nm). Cell lysates were immunoprecipitated with an anti-EAAC1 antibody, and the immune complexes were analyzed for PKCα immunoreactivity. Membranes were stripped and probed for EAAC1 immunoreactivity (WB: EAAC1). In the bottom panels, total cell lysates (25 μg of protein) were probed for PKCα immunoreactivity.
