MLN and peritoneal cells produce ROS and IL-1β induced by the ATP. MLN and cells obtained from peritoneal wash were incubated with H2DCFDA for 30 min at 37°C, washed and incubated for 15 min with 1 mM ATP. ATP-induced ROS production from Balb/c (a) and C57BL/6 WT and C57BL/6 P2X7−/− (b) mice was analyzed by flow cytometry. Cells from BALB/c mice were incubated or not incubated with 10 ng/ml LPS for 4 h at 37°C followed by incubation with 1 mM ATP for an additional 3 h.
