SDF-1 induces translocation of phosphorylated CREB into the nuclei of sympathetic neurons. E8 sympathetic neurons were cultured in defined minimal medium for 6 hr (A1A3) and then exposed to 100 ng/ml SDF-1 (B1B3), SDF-1 plus 20 μm CXCR4 antagonist AMD3100 (C1C3), SDF-1 plus 100 ng/ml pertussis toxin (D1D3), or SDF-1 plus 200 nm PKA inhibitor PKI (E1E3). After 30 min the cultures were fixed and stained for phosphorylated CREB (red) and Islet-1 (green). SDF-1 induces translocation of CREB into the nuclei of sympathetic neurons, and this translocation is blocked by all three pharmacological agents.
