Cardiac BTK is activated in CLP mice and reduced by ibrutinib or acalabrutinib. Mice were randomly assigned to undergo CLP or sham surgery. One hour later, mice were treated with ibrutinib (30 mg/kg i.v.), acalabrutinib (3 mg/kg i.v.), or vehicle (5% DMSO + 30% cyclodextrin i.v.). At 24 h after CLP surgery, the activation of BTK in the heart was analyzed by western blot analysis (n = 5 per group). Specifically, densitometric analysis of the bands is expressed as relative OD of (A) phosphorylation of BTK at Tyr223 corrected for the corresponding total BTK and normalized using the related sham band. (B) Phosphorylation of PLCγ at Tyr1217 corrected for the corresponding total PLCγ. All data are expressed as mean ± SEM for n number of observations. A value of ****P < 0.0001 was considered to be statistically significant when compared to the control by one-way ANOVA followed by a Bonferroni's post-hoc test.
