Pivotal role of Roquin during HCMV lytic production. (A) HFF cells were infected with HCMV Toledo (MOI = 2) and harvested at the indicated time points for immunoblot. HCMV was irradiated with UV-C (300 mJ/cm2) before infection. (BF) HFF cells were treated with siCon or siRoq, infected with HCMV (MOI = 2), and harvested at the indicated time points. (B and C) mRNA levels of Roquin (B) and viral genes IE2, UL44, and UL99 (C) were measured by qRT-PCR. (D) Protein levels of various viral proteins and Roquin were measured by immunoblot. (E) Viral genomic DNA levels were measured in cells transfected with two different siRNAs targeting Roquin. (F) Cell-free virus in the supernatant was titrated by limiting dilution analysis. Data represent mean ± SEM, n = 3; *P < 0.05 according to two-way ANOVA with Dunnett’s multiple comparisons test. (B, C, and E) Data represent mean ± SEM, n = 3; **P < 0.01; ***P < 0.001 according to two-tailed Student’s t test; NS, not significant.
