PCA of global gene expression in PBMCs stimulated with PMA, LPS, and ionomycin under equi-nicotine conditions. Human PBMCs were pre-treated with low, medium or high dose WS-CM (0.1, 1.0, and 3.0 µg/mL equi-nicotine units, respectively) or STE (100 µg/mL equi-nicotine units) for three hours followed by stimulation with the agonists (PMA [50 ng/mL], ionomycin [500 ng/mL], and LPS [1 μg/mL]) for five hours. PCA was carried out using R. Gene expression level of all probe sets was dimensionally-reduced and the first top three principal components were presented. While the samples pretreated with high- and medium- WS-CM are distinct and far separated from the control, those pretreated with low WS-CM and STE are more similar and clustered closer to the control samples.
