CENH3 Nucleosomes Are Highly Phased with Specific Sites on Monomers of the Wheat Centromeric Satellite CentT566 around WW Dinucleotides.
(A) Snapshot of the distribution of the wheat centromeric satellite repeat CentT566 occupied by CENH3 nucleosomes in two versions of the bread wheat reference genome (IWGSC_v1.0 and TGACv1). The horizontal scale marks the centromere locus (in kilobases) on each chromosome or scaffold.
(B) and (C) Distribution of the midpoints of CENH3 ChIP (B) and input (C) reads (100 to 200 bp) along the dimerized CentT566 satellite consensus sequence. Five major sites occupied by the CENH3 nucleosome position were observed in dimerized CentT566. The same nucleosome binding position is indicated in the input sample.
(D) Sequence features of CENH3 and canonical nucleosomal DNA. The dinucleotide frequency of WW and SS is displayed within ±200 bp from the center. The green curve represents input-WW, the blue curve represents CENH3-WW, the red curve represents CENH3-SS, and the purple curve represents input-SS. The x axis represents the distance from the center (bp); the y axis represents the frequency of dinucleotides.
(E) Dinucleotides frequencies associated with CentT566-related CENH3 nucleosomes. Blue curve represents WW; red curve represents SS.
