Complementation of the Atvip2 mutant by constitutive expression of AtVIP2. Three independent transgenic events were generated, and molecularly characterized before performing root transformation assays. (a) Roots of the wild-type, vip2 mutant and three transgenic Atvip2 lines expressing AtVIP2 cDNA were infected with a tumorigenic strain A. tumefaciens A208, at 1 × 107 CFU/ml concentration. Tumors incited on the roots were visualized and scored 4 weeks after infection. (b) Stable GUS expression. Roots of the wild-type, Atvip2 mutants and transgenic Atvip2 lines expressing AtVIP2 cDNA were inoculated with A. tumefaciens strain GV3101 carrying the uidA-intron gene within the T-DNA at 1 × 107 CFU/ml concentration. The inoculated roots were stained with X-Gluc 2-3 weeks post infection. All the experiments were repeated two times with similar results.
