Modular plasmids differing only in the firmicute replication module were introduced into H. modesticaldum by conjugal transfer from E. coli S17-1 harboring pBAD33 or pPB347. Conjugants were plated on mPYE plates supplemented with 10 μg/ml erythromycin.
