Depletion of ME31B increases ZIKV and WNV replication in Ae. aegypti cells. (A and B) Effect of ME31B, ATX2, and AAEL018126 (AAEL) silencing on viral RNA replication of ZIKV and WNV. At 6 h post-second dsRNA transfection, cells were infected with (A) ZIKV, ZIKV∆SF1, (B) WNV, or WNV∆SF1+2 at a multiplicity of infection (MOI) of 0.1. Viral genomic RNA (vgRNA) copies were determined at 48 h postinfection (hpi) by qRT-PCR normalized to the rpS7 reference gene and computed relative to the dsLuc transfected control. Statistics were performed by one-way ANOVA with Tukey post hoc test. (C and D) Effect of ME31B silencing on the virus growth kinetics of ZIKV and WNV. At 4 h post-second dsRNA transfection, cells were infected with (C) ZIKV, ZIKV∆SF1, (D) WNV, or WNV∆SF1+2 at an MOI of 0.1.
