Schematic diagram of the anti-influenza virus assay and cytotoxicity assay for TSN and OCN in MDCK cells. (A) In the pretreatment assay, MDCK cells were treated with various TSN concentrations for 6 h before infection with 10 MOI influenza A virus. In the cotreatment assay, 10 MOI influenza virus and various concentrations of TSN were first mixed and incubated at 4°C for 1 h before addition to MDCK cell cultures for 2 h. In the posttreatment assay, MDCK cells were first infected with influenza A virus for 2 h at 37°C and then treated with TSN for the indicated time. (B) The viability of MDCK cells was analyzed by MTS assay after 24-h treatment with the indicated concentrations of TSN or OCN (up to 10 µM) (TSN CC50 > 100).
