Induction of capacitative calcium (Ca²⁺) entry in MG‐63 cells. Fluo‐3‐loaded cells were treated with 5 µm thapsigargin (Tg) in Ca²⁺‐free HEPES‐buffered saline solution. After agonist‐mediated intracellular Ca²⁺ release, Ca²⁺ (final concentration of 1.8 mm, right arrow) were introduced into the buffer alone (○) or in the presence of (□) 2‐APB (100 µm) or () SKF‐96365 (30 µm). Each response shown is from at least three experiments with cumulative analysis of between 30 and 40 cells per field. Inset: addition of calcium to Ca²⁺‐free HEPES‐buffered saline solution without prior incubation with Tg.
