Nuclear localization signal (NLS) is critical for activity of an effector domain fused to C-terminus of dCas9. An additional nucleoplasmin NP-NLS appended to the C-terminus of the catalytic domain of DNMT3A clearly showed an increase in activity of the dCas9 fusion (solid lines) compared to activity obtained using the dCas9 fusions without additional NP-NLS (dotted lines).
