Stimulation of IL-6 gene transcription by membrane depolarization depends on Ca2+ influx and activation of CaM-dependent protein kinases. PC-12 cells were transfected with a reporter gene construct containing the luciferase sequence as reporter and the human IL-6 promoter (−1179/+9). Cells were stimulated by 45 mm KCl for 6 hr. Inhibitors were added 1 hr before stimulation. Data are mean ± SEM values of three to four independent experiments, each done in duplicate. *p< 0.01 compared with untreated control. +p < 0.05 compared with cells stimulated with 45 mm KCl.
